Force measurements are obtained through observation of the Hookean bending of the glass needles, which are calibrated by a direct and empirical method3. Equipment requirements and procedures for fabricating, calibrating, treating, and using the needles on cells are fully described. The force regimes previously used and different cell types to which these techniques have been applied demonstrate the flexibility of the methodology and are given as examples for future investigation4-6. The technical advantages are the continuous 'visualization' of the forces produced by the manipulations and the ability to directly intervene in a variety of cellular events. This video playfully demonstrates the versatile manipulation of single biomolecules using JPK's NanoTracker optical tweezers platform, simultaneous to sensitive fluorescence microscopy in a JPK's microfluidics flow cell. A native (non-fluorescent) DNA molecule is suspended between two optically trapped beads. A continuous fluid flow is applied using a multichannel laminar flow cell. The buffers flowing by can be changed by moving the microscope sample stage between various laminar channels. In the video, the trap-held DNA molecule is suspended successively in a plain buffer and in a flow of fluorescently labeled DNA molecules, coated with fluorescent proteins that specifically bind to the DNA. Towards the end of the video, one such fluorescent DNA molecule connects to the invisible one, like a rope on the washing line. Such assays can be used to combine quantitative mechanical manipulation using optical traps with protein binding kinetics studies using fluorescence. In 1966 he completed research on the subject and proposed the concept of relation between spinal mechanical disturbances and development of spinal dystrophic diseases. He was searching for the way to improve performances of professional athletes, prevent spinal column sports related injuries and disproportional to the young age development of dystrophic disease in spine by improvement of spinal bio-mechanics. He called his methodology clinical bio-mechanics/cranio-sacral therapy. During his study he found out that spinal movement segments that are repeating themselves and creating entire spine are related from the prospective of bio-mechanics. Mobile humanoid robots must be able to maneuver in irregular terrains while performing accurate physical interactions with the environment and with human operators. These types of capabilities entail developing new dynamical models that can adapt to the terrain while creating controllers that can stabilize the robot by balancing on its wheels. In particular a description of the robot that includes underactuated free-body dynamics with contact constraints between the wheels and the terrain is considered. Moreover, our humanoid robot incorporates a biarticular mechanical transmission that we model as a force constraint. Using these tools, we develop new compliant controllers that can endow the necessary multiobjective skills to maneuver and balance in the terrains while stabilizing the self motions of the highly redundant robot. To validate the theoretical developments, we build a new mobile humanoid robot that incorporates an upper body torso-arm-head system, and a new torque-controlled holonomic base developed by our laboratory. Various experiments are conducted, including balancing using feedback on the robot's center of mass and maneuvering in an irregular terrain while interacting compliantly with a human user.. + Informações
